Anti-HLA I Antibodies Induce VEGF Production, Increasing Endothelial Proliferation and Paracellular Permeability
Researchers at the University of Bern in Switzerland have discovered that anti-human leukocyte antigen class I (HLA I) antibodies can activate several protein kinases in endothelial cells, leading to increased proliferation and cell survival. This finding may provide insight into the underlying mechanisms of antibody-mediated rejection and the formation of interstitial edema. The study found that anti-HLA I antibodies induced the production of vascular endothelial growth factor (VEGF) by endothelial cells, which activated VEGF receptor 2 (VEGFR2) in an autocrine manner. This led to a c-Src-dependent phosphorylation of vascular endothelial (VE)-cadherin and its degradation, resulting in impaired adherens junctions and increased endothelial permeability.
