Modulation of Cellular Redox State Underlies Antagonism between Oxaliplatin and Cetuximab in Human Colorectal Cancer Cell Lines
Recent research published in the British Journal of Pharmacology has found that the efficacy of oxaliplatin, a platinum-based compound effective in treating colorectal cancer, can be impaired by the presence of the cetuximab monoclonal antibody. The study, conducted by L. Dahan and colleagues at Aix-Marseille University, evaluated the effect of cetuximab on the efficacy of oxaliplatin in a panel of colorectal tumor cell lines. The researchers found that combination of oxaliplatin and cetuximab was less cytotoxic than oxaliplatin alone in cells with wild-type Ras, but cetuximab did not affect oxaliplatin efficiency in cells with K-Ras(V12) mutation. The study suggests that the efficacy of oxaliplatin relies on the endogenous production of H2O2, and that cetuximab impairs oxaliplatin efficiency by affecting the redox status of cancer cells through the EGFR/Nox1 NADPH oxidase pathway.
